- filter the solution through a Whatman No. 1 filter to remove any particulate matter

- Destain for Coomassie gel is the same as above but without the dye

Fix solution

add dH2O to 10 ml

- store @ 4o C for up to a week

- working soln: 950µl Fix + 50µl formeldehyde


10% Formaldehyde

ddH2O 70-80 ml
Paraformaldehyde 10 g
NaOH 1 pellet

- stir for 30min-1hr until soln is clear (if it remains cloudy use newer formaldehyde)

- pH to 7.2-7.4 using conc HCl

- bring up to 100 ml - store @ 4o C for up to a month

Stripping Buffer for Western Blot

  add: [Final]
Tris pH 7.0 31.25ml of 1.0M Tris pH7.0 62.5mM
ß-ME 3.41ml 100mM
SDS 100ml of 10%SDS 2%
adjust volume to: 500ml  

- To strip blot incubate at 50° C for 30 min with occasional shacking
